It can easily be done if it is a revocable trust. If it is an irrevocable trust, it will require the written consent of the present beneficiary (I assume you), the trustee (you), and the remainder beneficiaries.

Call the clerk's office in the county where he died and get the phone number of the estate's lawyer. He will probably talk with you. If that doesn't work, go down to the probate clerk's office yourself and ask to see the file. The file will contain a will (if there was one) an inventory of assets and any claims. If it looks like your sister is not recognizing you as a beneficiary and is boxing you out, you can either hire a lawyer or write a letter to the judge and complain. The judge will most likely call a hearing and invite you to attend.

Yes, that is one way that you can be enabled to deal with her financial affairs. Another way is for your mother to enter into a fully funded revocable living trust naming you as successor trustee. For many reasons this is a better way of dealing with incapacity, although most people probably use the durable power of attorney. Having said that, if your mother grants you that power, make sure that it is the new version of the durable power of attorney, as the statute in Florida was significantly changed in October of 2011.
