If you brother files for you, it takes a minimum of 10 years before you are eligible to adjust status. You can only adjust if you maintain your status in the US. Your brother petitioning for you does not give you the eligibility to be in status, you will have to find other means to renew your permit or apply for an H1b visa, etc.

If you filed an I-130 visa petition for your wife, that should have been approved within 6 months. If she entered the US without inspection, she cannot adjust in the US and must go home for an immigrant visa. When she can reenter the US will depend on many factors and that is probably why she is still waiting.
