Can we apply for one first, and once approved, can that person be the co-sponsor for the affidavit of support of the second applicant?

The co-sponsor will have to disclose on the second affidavit of support how many immigrants he/she sponsored in the last 5 years; these previously sponsored immigrants are counted as members of the co-sponsor's household. If the co-sponsor's income is not enough to support 2 immigrants at once, it will not be enough to support the second one after the first gets approved - unless the affidavit of support for the second immigrant will be filed 5 years later.

Probably the best thing you can do is to write to the applicable USCIS field office where the interview will be conducted or the US Consulate if the interview is to be overseas giving as complete details as you can concerning the intending immigrant and the petitioner. You may also wish to send your correspondence by certified return receipt to keep a record of when the appropriate agency received your withdrawal notice. He can submit the I-130 and even get it approved, but he will need to get a J-1 waiver in order to get permanent residence.
